Project

Piezo1 is a cation channel that allows calcium influx and initiation of downstream calcium signaling events. It is abundantly expressed in primary and MLO-Y4 osteocytes, the major mechanosensing cells in bone. In vivo injection of Piezo1 activator, Yoda1, was previously show to promote bone formation in mice. However, whether the administration of Yoda1 will enhance osteocyte mechnosensitivity and decrease breast cancer cell migration are still unclear. Using our previous developed microfluidic platform that allows bi-directional cellular signaling and cancer cell extravasation, I will focus on investigating the effect of Yoda1 on osteocytes mechanosensitivity and breast cancer growth reduction. This will hopefully provide an insight into rescuing aged bone cell mechanoresponse and maintaining bone integrity for elderly breast cancer bone metastasis patients
